Severn Biotech now supplies IPG strips
for 2-D gel Electrophoresis
Now available in different pH gradients and differing lengths of strip to suit mini gels and larger slab
gels. The Severn Biotech IPG strips are inexpensive and accurate in mobilisation of proteins for IEF.
The strips are simple to use, the direction is denoted by the sharp end of the strip, they are simply re-
hydrated overnight either passively or actively. The strips are now ready to focus by electrophoresis.
Simply peel off the strip from the blunt end (cathodic) and place in your first dimension equipment.
Check the strip is evenly covered in running buffer and is pointing towards, the anode.
If cup loading is to be used, the strips must be rehydrated in the appropriate buffer and then the
sample loaded using a suitable cup holder.
The rehydration volume for 7cm strips is 125 µl and sample loading is recommended at 4-8 µg. for
silver staining and 10-60 µg. for Coomassie staining. However optimum protein loading should be
determined for a particular sample. Focus the proteins at a maximum of 50 mA per strip. The
focussing parameters are; 500v for 30 min, 1000V for 30 min and 5000V for 1h 40 min. This is a
recommended starting protocol and the optimum parameters for focusing should be determined for
your sample.
IPG Strips are available in different sizes and we presently manufacture a number of different pH
gradients.
Severn IPG Strips are available in 7cm,11cm,17cm 21cm and 24cm sizes.
Other sizes can be made as bespoke IPG strips if requested, so please enquire.

Storage
IPG strips should be stored at –20C and it is recommended to avoid frequent freeze/thawing.
Expected shelf life is 12 months from date of purchase.
Shipping
All IPG strips are shipped frozen under dry ice on a next day, courier basis and not delivered or shipped
on Fridays to avoid thawing. Please freeze as soon as received.

launched Severn
Super Sequencer, a new
Acrylamide/Bis-Acrylamide mix containing a new novel
proprietory cross linker. The new sequencer gives a longer read length enabling the researcher to get the most out of an acrylamide gel thus giving a cost saving as well as a read advantage to both manual and automated sequencing. The new solution has been designed to work with the ABI 377 Prism DNA Sequencer using Sanger Di-deoxy dye termination sequencing . The new Severn Super Sequencer can give read lengths of over 800 bases in length when used with automated sequencers .The minimum read for QC requirements are 300 bases when tested on manual systems. Severn Super Sequencer is available in two different formulations for use in sequencing and gene scan work, they are 6.5% and 5% respectively. Both formulations are supplied in 3 different pack sizes namely :- 500ml , 1000ml and for the busy researcher 5000ml. Catalogue Numbers
RNase Free Water (See Other Water products listed Below) New sterile products include PCR water, available in 5ml vials for the discerning molecular biologist who requires Ultra purified water in a sterile form, free from contamination from other users. This product is in an easy to use 5ml pack autoclaved and bacterial filtered and treated with DEPC to give peace of mind in valuable PCR reactions with Taq Polymerase.
Ex-tract DNA/RNA Extraction Kit Extract is a new Kit from Severn Biotech comprising three reagents for the extraction of RNA and DNA from whole blood , biological fluids et al. The kit is designed for total extraction using chaotropic agents and an extraction Matrix to capture the Nuclear material from solution by solid phase binding. The extract is then eluted from the matrix and concentrated.
SYBR Dyes SYBR Green I and II are now available from Severn . These dyes are used exclusively in staining gels for DNA . They are highly sensitive (60pg ds DNA using a 1:10,000 dilution of SYBR dye) and a safer alternative to Ethidium Bromide. Sensitivity is increased with when used in conjunction with Yellow Green Filters . They do not affect the activity of restriction endonucleases at typical staining concentrations .
Orange G Severn Biotech now manufactures a number of Dyes in Solution in a Ready to Use (RTU) format. The latest of these is Orange G in a 1ml solution for use in PCR fragment analysis . Orange G ( 7,hydroxyl-8- phenyl azo 1,3, napthalene disulphonuic acid) runs faster in DNA gels than bromophenol blue and is therefore used as a forward marker in PAGE for SNP and PCR analysis . The dye is available in 1ml tubes and available in single tube packs either made up in buffer or Purified water . Other dye stains in the range include Bromo-phenol Blue Loading dye available in 10ml packs incorporating xylene cyanol and sucrose , Ethidium Bromide Solution in 10mg/ml concn available in 10ml packs and SYBR Green I and II in 500microlitre packs .
Cryo-Vials Severn Biotech Ltd is now supplying cryogenic vials, storage equipment and microtubes for cryopreservation and PCR. The range is designed for cryostorage of human, animal and biological fluids, for example, blood, serum, sperm, bacteria and in fact most biological substances. The cap and vial are designed to fasten without the use of gaskets and are made from polypropylene devoid of chemical softeners and cadmium. This gives better sealing and avoids the different expansion rates seen when caps and vials are made from different materials. A unique feature is a star base foot which, when used with a cryorack, it locks the vial in position to give ease of use, with one handed operation. Cryovials can be stored in liquid nitrogen and are available in different sizes of volumes and quantity from 0.2ml PCR microtubes to 50ml Blood sep filter tubes. Tubes are sterilised by irradiation.
