This bombesin analogue has been found to be a high affinity ligand for the human orphan receptor, bombesin receptor subtype 3 (BRS-3). Neither bombesin itself nor any of its analogues that bind to other bombesin receptor subtypes exhibited significant affinity for BRS-3. Binding of this peptide to BRS-3 stimulates the formation of inositol phosphates. Guanosine 5'-(ß,-imido)-triphosphate causes a change in receptor affinity, preventing binding of this ligand. These results indicate that for a member of the bombesin receptor family, BRS-3 has a unique pharmacological profile. The natural ligand for the BRS-3 receptor subtype is presumably either an undiscovered member of the bombesin family or an unrelated peptide.

homogenates. These results suggest that this analogue will be valuable in assessing the functional relevance of

Urocortin binds more effectively to CRF Type 2 receptors than CRF itself and is probably the endogenous ligand.

Selective FSH-releasing activity has been demonstrated on a number of occasions, but until now the molecule responsible for this action has not been isolated. In view of the FSH-releasing action of LHRH, this peptide has often been postulated to be responsible for both FSH and LH secretion from the pituitary. Recent in vitro and in vivo experiments with rats have shown that lamprey LHRH-III selectively stimulates the release of FSH without affecting LH. It is postulated that this sequence may represent the long sought after endogenous FSH-releasing factor.

PrRP-31 and PrRP-20 are the first reported hypothalamic peptides that exclusively stimulate the release of prolactin from the pituitary without affecting that of other known hypophyseal hormones. Interestingly, the discovery of these peptides is an example of an orphan peptide receptor being used to identify its own ligand. Working backwards from the human orphan receptor, hGR3, Hinuma et al. (1998) identified a peptide ligand in bovine hypothalamic extracts. After isolation of the respective cDNA in bovine brain, they were able to identify the 31-amino acid peptide (PrRP-31) and 20-amino acid fragment (PrRP-20) corresponding to this sequence. The peptide sequences are reported to be highly conserved in other species (human, rat). PrRP-31 has been shown to stimulate prolactin release from a rat pituitary RC-4B/C adenoma cells, from anterior pituitary cells from lactating rats, and from rat pituitary cells in a perfusion assay.

This analogue represents one of a new class of pharmacological tools that can be used for investigating the complex cardiovascular actions of AVP and may form the basis for the development of novel antihypertensive agents.

Storage

 All peptides are supplied lyophilised to avoid degradation of the product. Most peptides if stored tightly sealed and at <-20ºC will remain stable for many years. Some peptides are stable at room temperature but it is difficult to predict so caution is sensible.

 To maintain stability it is recommended that before opening, vials are warmed to room temperature in a desiccator with fresh desiccant to prevent condensation forming on the product. However, water adsorption is unavoidable for some hygroscopic or deliquescent peptides.

The appearance of a peptide is usually a fluffy colourless powder but this can vary in density. Some peptides are freeze-dried to a very dense tar like form, (Usually still colourless). Others form oils. Some disappear as soon as the vial is opened, as they are deliquescent. Simply put it is impossible to judge the weight of a peptide from its appearance.

 Once aliquoted peptides should be in a buffered solution pH5-7 in sterile purified water. These should be stored at <-20ºC and once reopened and thawed any remaining peptide in an aliquot should be discarded.

 Dissolving 

Some peptides can be very difficult to dissolve in pure water. The following measures may be taken to aid dissolution:

-          Sonication to break up particles to speed up dissolution.

-          A small amount of DMF or DMSO may be used to dissolve the peptide and then dilute with water.

-          Most peptides are soluble in 67% aqueous acetic acid. The exceptions are usually very acidic and in these cases aqueous ammonia solution usually works.

-          Addition of a chaotrope such as urea or guanidinium hydrochloride may aid dissolution of some peptides. These can interfere with biological systems so should be used sparingly.

 Biological Activity

 We provide a peptide with the correct amino-acid sequence, mass and purity. This does not necessarily confer the correct biological activity. Biological activity is affected by a number of factors.

The peptide must have the correct conformation. This can be affected by the pH, the solvent used, trace impurities, salts, the lack of a co-factor and even the reconstitution method.

 For example HIV Protease showed a greatly reduced biological activity on lyophilisation and redissolution in water. After much experimenting the following method was devised:

-          Dissolve at 5mg/ml in 50mM sodium acetate, pH 5.5 containing 8M urea, 1M EDTA and 2.5mM dithiothreitol.

-          To refold dilute with 9 volumes of 50mM sodium acetate pH5.5 containing 1mM EDTA and 2.5mM dithiothreitol, 10% glycerol, 5% ethylene glycol and 0.2% Non-idet P-40 at 4ºC.

-          Full specific activity returned.

 This is an extreme case but it is worth considering this factor if there is a reduced activity.

Peptides are supplied as a salt of triflouroacetic acid and sometimes acetic acid/ammonia. As a result most have a pH of ~2-3. It is therefore necessary to neutralise the peptide and thoroughly check the pH before use as some peptides show a massive change in activity at altered pH. N.B. some peptides may have been neutralised by us if they contain an acid labile sequence, (e.g. Asp-Pro).

Another important factor with pH is that if a peptide contains a cysteine in the free thiol form dimerisation can occur. This happens at its greatest rate at pH7.8. As this is in the range used for many tests rapidity is essential.

In some systems the ions themselves can play a role, acetate is part of many metabolic pathways after all. If this is suspected the peptide should be desalted by passing through a desalting column such as our Super Desalting Column Plus.

NOTE: WE PROVIDE A MOLECULE TO GIVEN SPECIFICATIONS. HOWEVER, EVERY PEPTIDE IS UNIQUE SO OPTIMISATION OF BIOLOGICAL ACTIVITY CAN ONLY BE DONE EXPERIMENTALLY BY THE CUSTOMER.
