Description

Solutions of Ethidium Bromide require removal once they have been applied to the gel for staining.  The excess material in the running buffer solution e.g TBE, TAE, MOPS can be removed by two distinctly different methods.

The first is removal of small quantities by adsorbtion onto a resin contained within a de-staining bag structure and commonly known as the T-bag method.This will remove approximately 5mg of ethidium bromide from solution per T- bag used.

The second method is removal by the use of activated charcoal carbon filter with a glass fibre pre-filter, within a polypropylene housing.  They are used to filter litre quantities of buffer and can remove greater quantities of Ethidium Bromide from solution compared to the T-bag method. i.e >100mg

The filter comes as a single unit and is used in conjunction with a side arm conical flask and water vacuum pump.  The filter will easily remove Ethidium from more than 10-20 litres of buffer.  The longevity of the filter can be assessed by shining a UV light at the filtrate, which will flouresce once the filter is exhausted.

Quanitification for both methods is see using a UV (spec) source with  wavelength of 285nm to indicate the absorbance of Ethidium Bromide.  This absorbance reading reduces as the Ethidium Bromide is removed from solution.
